Hi, I have a question on how to calculate the Steady State accumulation index in a different method than the one in the WinNonlin NCA object. Is it possible to calculate the actual Accumulation ratio i.e. AUC(tau,ss)/AUC(tau,day1) in Phoenix ?

Hi there, this is indeed possible and something we cover in our "Introduction to Phoenix WinNonlin course". Here are some simple steps to calculate these sort of ratios once you have executed the NCA.

1) if not already in separate sheets - use the SPLIT function on Day to give the an Output Parameter sheet per day.

2) Merge the two sheets back together (you may choose to drop those columns you are no longer interested in)

3) with the AUC values of Day 1 and your steady-state day on the same row you can now simply perform your calculation using a column transformation within a DataWizard object.

I hope that makes sense. Simon.
