Hi,

 

I have a study with two time dosing per day.  the bleeding was 0.5, 1, 2, 4, 8 (before the second dose), 9, 16 and 24 post first dose.  This means we have 1, 8 hours post second dose.

 

So is it accurate to say  that the area under the curve after the second dose is over or under estimated.  Please let me know what you think.

Your study design isn't really appropriate for NCA analysis,  which can really only deal with single naive dose or a dosing interval at steady state.

Your period from 8hours onward is not really likely to be at steady-state and is not really sufficiently sampled to estimate the AUC. Depending what method you use, (linear trapezoids throughout, or lin up, log down you might over or underestimate the profile, esepcially depending on where Tmax is tending to occur.

I would recommend trying to fit the data with a compartmental model if you want to estimate these two AUCs better.
