Hello, I want to build a pharmacokinetic model for methotrexate, but I am not familiar with using pharmacokinetics software, can anyone point me a direction please?
So, what I want to do here is:
Methotrexate is activated into methotrexate polyglutamate form inside the red blood cells. I want to see if patients were on parenteral form (especially subcutaneous route) of methotrexate, and they have achieved steady state (I mean steady state of methotrexate polyglutamate inside the red blood cells). What I am trying to find out here is: I want to build a PK model to simulate if the patient skip a dose of parenteral methotrexate, how does it affect the concentration of methotrexate polyglutamate inside the red blood cells.

I hope anyone could help me with this, even if it is just a rough guide, it will be really appreciated. Thanks in advance.

You could use RsNLME (population pharmacokinetic) for building your model. If you are an academic user RsNLME is free and we can help you to set it up and run the model. For non-academic we can offer 4 weeks trial license.

You could use RsNLME (population pharmacokinetic) for building your model. If you are an academic user RsNLME is free and we can help you to set it up and run the model. For non-academic we can offer 4 weeks trial license.

Are you thinking something like this? a basic 2 com model plus intracelluar kientics of RBC

We also have attractive terms for academia for Phoenix, generally missing a dose whilst at steady state has little overall impact on the PK;

Exactly, something like that, but I am curious to find more about that, not just one dose, maybe 2 or more. Not too sure if that can be done on other software, like Monolix, do you have any experience in this by any chance?

Yes, we could design using RsNLME or Phoenix NLME. Please refere to PML School on Certara Forum and hope this will give you a better Idea what you could acheive with RsNLME/Phoenix NLME.
