Any chance you can share anything like the input data or the project file with us? If this is confidential data, you might want to reach out to support@certara.com for help.

Also if you haven't already, I would suggest to watch the tutorial under NLME linked from the the help menu and/or followed the examples provided in the help;

https://onlinehelp.c...el_examples.htm

that explain the file found in; C:\Program Files (x86)\Certara\Phoenix\application\Examples

Thanks, Bernd,  for your prompt reply.  Yes, I have data as follows:

Route IV Bolus (1mg/kg)     IV Infusion(18 mg/kg/h*0.5 h) Dosing Solution 10 mg/kg   time Concentration Time after bolus/or infusion stops Time after bolus dosing(hr) (ng/ml) 0 0 / after IV Bolus-3 min 0.05 16767 IV Infusion-15 min 0.3 44367 IV Infusion-30 min 0.55 59600 2 min 0.58 61500 5 min 0.63 50967 10 min 0.72 33167 15 min 0.8 23000 30 min 1.1 5230 45 min 1.3 1490 1 h 1.6 771.3 2 h 2.6 293.0 4 h 4.6 100.2 6 h 6.6 69.33 8 h 8.6 44.20 24 h 25 /

Please note: The first column indicated the time after IV bolus was given (0 h) and 3 min later infusion started; IV infusion-15/30 min: 15 and 30 min into the infusion;  2 min-24 h: time after infusion stops.

Sorry, this data format does not work. Let me find a different way.

Hi, Simon:  Here attached is the PHXPROJ file. Let me know if you have any questions regarding the data.

Hi Yuan,

    Please find attached your project with a few models added,  It looks like it maybe tricky with this data to have good confidence on the model selected and you may want to also look at others, maybe TMDD is appropriate.  I have also used and append to show how to bring all the data into one work sheet if you want to model it together,

    However the bolus and Infusion data looks quite different in terms of clearance than the other two, so if it is the same compound then that is another challenge, of course if you have more data that will probably help.

    Setting up the dosing can be done in an "internal" worksheet in Phoenix which is like a scratchpad within the model but it's often better practice to have your doses in your data.  I think I transcibed them correctly from your earlier post, but I suggest you double check them note I have put them in the same units as your concentration so it's easier to check your output units;

Thanks a lot, Simon. I will take a close look at what you have done and figue out how to do it the next time. Thanks again.  Dan
