I would like to simulate an oral PK profile with 3-compartment disposition, I'm trying to use the PK modeller as attached. Can you get it to run? Thanks, Charlie [file name=simulate.phxproj size=98049]http://www.pharsight.com/extranet/media/kunena/attachments/legacy/files/simulate.phxproj[/file]

Hi Charlie, here you go, I attach the updated project plus the screen shots beliw show what i changed to get the simulation to run. missingC.jpg Don't worry when I first started using the Phx model I kept forgetting to complete this since it's already filled with 1000 points in the WNL classic engine, but if you give some values like below then you will get your result; [file name=simulate-20130621.phxproj size=350093]http://www.pharsight.com/extranet/media/kunena/attachments/legacy/files/simulate-20130621.phxproj[/file] sim-20130621.jpg Simon. PS there you go two answers at the same time!

Dear Charlie I modified your project to enable simulation. Please take a look and let me know if it is clear. You had to add a column for the response and also write C as the response you want to simulate. Adding a column for DV may be not a must but I did it. best Serge [file name=simulate_official.phxproj size=489176]http://www.pharsight.com/extranet/media/kunena/attachments/legacy/files/simulate_official.phxproj[/file]

Thank you Simon and Serge! Please could you also let me know where I've gone wrong in trying to use the macro model? Charlie [file name=simulate_macro.phxproj size=806303]http://www.pharsight.com/extranet/media/kunena/attachments/legacy/files/simulate_macro.phxproj[/file]

tricky one this Charlie, I couldn't see anything immediately wrong but on checking the release notes I found a known issue. QC_PHX10246

In Individual modeling, Simulation mode, with macro parameter models, the Y variable defaults to C, when it should be C1. The user has to change it to C1 to get meaningful output

Sorry about that,

Simon. Posted Image

Please could you help again? Can you explain what's happening to the extravascular macro model? The extravascular micro model seems to work OK. Thanks, Charlie [file name=simulate_macro-20130624.phxproj size=1310630]http://www.pharsight.com/extranet/media/kunena/attachments/legacy/files/simulate_macro-20130624.phxproj[/file]

Hi Charlie, using the initial estimate tool it looked like your kA was perhaps a factor of 100 out?

If I increase it to 7 then the fit looks OK and we get the following;

Does that seem feasible?

Simon.

Hi Simon,

The input half-life is predicted to be approx. 10 h, so Ka is 0.07 /h.

This value seems to work OK for the micro-constant model so why do I get a different profile with the macro-constant model?

Regards,

Charlie
