non-compartmental Analysis vs.compartmental

When is it appropriate to use non--compartmental analysis? We are testing new compounds in an SAR series for better stability. Our PK is IV bolus only, followed by sampling out to 8 hrs. Our analytical chemist uses compartmental analysis in WinNonLin to determine PK parameters. Is this appropriate or should it be non-compartmental?

Dear user,

It would depend on your objective and your design. If your goal is to determine PK parameters without the intention to do further simulations and you have an appropriate design then NCA might be appropriate. If you have a rich design that expands the terminal phase your NCA determinations and Compartmental determinations will probably be very similar.
