therapeutic concentration window and AUC

I have plasma concentration time values for a number of subjects following a single dose: I have a feeling for what the therapeutic window is. I believe it is between 3 and 15 ng/ml , so that, if the concentration is between this window, then we have efficacy. I am wondering if the program can calculate the times corresponding to these values and then calculate AUC. By this strategy we can calculate the AUC for the therapeutic window. I have the AUC to inf so then we would know the percent AUC for the therapeutic window.

Angus - this is very easy and built into NCA (since at least version 5 of WNL).

Simply choose internal worksheet on the option "therapeutic response" and enter your values there; e.g. below I chose to uncheck all sort keys when building the worksheet.

you will then automatically get time and AUCs spent above, below and within these boundaries

the Min and Max columns are displaying the min and max conc's seen in the actual data, so obviously in my screenshot the proposed therapeutic window would be much less than the observed data

Thank you Simon I see it there! For many years I used another program for that type of calculation. Never have I used WinNonlin for that kind of calculation. It looks interesting.

 

I used to use WinNonlin for compartment model simulations.
