Calculate AUC/MIC, Cmax/MIC and %T>MIC using Phoenix

Hello everyone,

 

I have conc-time profile from multiple dose study of an antibiotic in mice and I am interested in calculating AUC 0-168/MIC, Cmax/MIC and %T>MIC. The MIC is 1600 ng/ml.

 

I have attached two excel sheets:

1. Conc-time profile

2. Dosing information

 

I would appreciate if someone can help me how to perform this using Phoenix Winnonlin .

Hi Chetan, this is pretty simple if you use the Therapeutic response option in NCA and you want to calculate this over the whole 168h profile.

 

1) set up the Lower limit to 1600, the resulting TimeHgh will be time spent above that level.

 

2) Send the resulting Final Parameters Pivoted to a DataWizard and create the three formulas you need.

 

remember the other NCA output e.g. clearance will not necessarily be valid as you have treated all the dose events as a single profile to calculate the AUC0-168 etc.

 

  In future it would probably be helpful if you started off the project so others could see what you had tried.
