Hi!

I am wondering if there is any way to calculate the SE for AUC(0-inf) when one is using the NCA module applied to sparse samples (destructive sampling).

Thanks,

Dora

Dora,

Since the standard error of the mean AUC is calculated based on linearity of the AUC function (stepwise function) as described in Nedelman and Jia (1998) and Holder (2001); it would be incorrect to apply this theory to the extrapolated portion of AUC.

We are however researching options (qc4858) to enhance this function and welcome any references users can suggest regarding SE calculation on logarithmic trapezoids.

Holder (2001). Comments on Nedelman and Jia's extension of Satterthwaite's approximation applied to pharmacokinetics. J Biopharm Stat 11(1-2):75-9. "

Hi Simon,

have a look at R's package PK by Martin Wolfsegger and Thoma Jaki. In the documentation you find references to the theoretical background of BE assessment based on AUCinf in serial sampling designs.
 Best regards,
Helmut
