Hi all,

There are totally four methods to calculate AUC in NCA. Normally I use linear up log down for PK data and linear/linear for PD data. Am I using the right methods? For what situation we need to use the rest two methods? Would you please provide more information on the application of these four methods?

Thank you and happy holiday!

LLLi, that sounds reasonable to me, there may be some PD response that would work better on a log scale e.g. bacterial growth but generally that should be fine..

The other methods support users who need these methods for their SOPs or backward compatibility with older analyses, e.g in my experience, some guidances originally said linear trapezoids since they were easier to calculate at the time and then with the general conservative nature of the pharmaceutical industry these have remained as defaults for some groups.

Happy New Year

  Simon.
